Apalutamide The mechanism of interaction is CYP3A4 Co-administration with REYATAZ

inhibition by atazanavir/ritonavir. contraindicated due to the potential
for decreased atazanavir and

subsequent loss of virologic
response and possible resistance to

section 4.3). In addition, serum
concentrations of apalutamide may
be increased when coadministered
with atazanavir/ritonavir, resulting
in the potential for serious adverse
events including seizure.
Encorafenib The mechanism of interaction is CYP3A4 Co-administration of encorafenib
inhibition by atazanavir and/or ritonavir. with REYATAZ (with or without
ritonavir) is contraindicated due to
the potential for loss of virologic
response, development of
resistance, increase in encorafenib
plasma concentration and
subsequent risk of serious adverse
events such as QT interval
prolongation (see section 4.3).
Ivosidenib The mechanism of interaction is CYP3A4 Co-administration of ivosidenib

Co-administration of Tyverb with irinotecan (when administered as part of the FOLFIRI regimen)
resulted in an approximate 40% increase in the AUC of SN-38, the active metabolite of irinotecan.
The precise mechanism of this interaction is unknown, but it is assumed to be due to inhibition of one
or more transport proteins by lapatinib. Adverse reactions should be carefully monitored if Tyverb is
co-administered with irinotecan, and a reduction in the dose of irinotecan should be considered.

Pazopanib is an inhibitor of the uridine diphosphoglucuronosyl-transferase 1A1 (UGT1A1) enzyme in
vitro. The active metabolite of irinotecan, SN-38, is a substrate for OATP1B1 and UGT1A1.
Co-administration of pazopanib 400 mg once daily with cetuximab 250 mg/m2 and irinotecan
150 mg/m2 resulted in an approximately 20% increase in systemic exposure to SN-38. Pazopanib may
have a greater impact on SN-38 disposition in subjects with the UGT1A1*28 polymorphism relative to
subjects with the wild-type allele. However, the UGT1A1 genotype was not always predictive of the
effect of pazopanib on SN-38 disposition. Care should be taken when pazopanib is co-administered
with substrates of UGT1A1.

In vitro studies indicated that crizotinib is a weak inhibitor of uridine diphosphate

UGT1A1 and UGT1A9 substrates
In vitro data indicate that regorafenib as well as its active metabolite M-2 inhibit glucuronidation
mediated by UGT1A1 and UGT1A9 whereas M-5 only inhibits UGT1A1 at concentrations which are
achieved in vivo at steady state. Administration of regorafenib with a 5-day break prior to
administration of irinotecan resulted in an increase of approximately 44% in AUC of SN-38, a
substrate of UGT1A1 and an active metabolite of irinotecan. An increase in AUC of irinotecan of
approximately 28% was also observed. This indicates that co-administration of regorafenib may
increase systemic exposure to UGT1A1 and UGT1A9 substrates.
